Abstract:
Targeted proteomics has emerged as a powerful protein quantification tool in biomedical research and systems biology due to many attractive features that cannot be provided by antibody-based assays. However, it lacks sufficient sensitivity to measure low abundant but functionally important proteins and posttranslational modifications in human bodily fluids or tissues. To address this issue our team has been focusing on technology development for significantly enhancing targeted proteomics sensitivity without the use of any specific affinity reagents. We have made several major breakthroughs in sensitivity for enabling reproducible quantification of low pg/mL levels of proteins in human blood and single-digit protein copies per cell in human cells. Such sensitivity is comparable to or even better than that of most analytically validated ELISA assays. The new targeted proteomics capability has been broadly applied in many biomedical projects as well as systems biology studies to answer biology questions and generate new hypotheses. In the talk new technology development and broad applications will be discussed and future perspective of targeted proteomics will be briefly mentioned.
